摘要
目的:观察针药结合对卵巢储备功能不足(decreased ovarian reserve,DOR)大鼠卵巢细胞凋亡因子和生殖激素的影响,探讨其作用机理。方法:将72只SD大鼠随机分为对照组、中药组、针刺组、模型组、西药组和针药组,每组12只。除对照组采用等量生理盐水灌胃外,其余五组均采用雷公藤多苷药液灌胃,建立符合实验要求的DOR大鼠模型。针药组、中药组予护卵汤灌胃,针刺组和模型组采用等量生理盐水灌胃,西药组采用克龄蒙药液灌胃,同时对针药组、针刺组进行针刺,以上操作持续16 d。采用ElISA法检测血清雌二醇(estradiol,E2)、促卵泡生成素(follicle-stimulating hormone,FSH)、黄体生成素(luteinizing hormone,LH)及抗缪勒氏管激素(Anti-Mullerian Hormone,AMH)水平,PCR检测卵巢组织中细胞凋亡因子Bax、Caspase-3、Bcl-2的表达。结果:与对照组比较,模型组FSH、LH、Bax、Caspase-3表达明显升高,E2、AMH、Bcl-2明显降低(P <0. 05);与模型组比较,针刺组、中药组、针药组及西药组FSH、Bax、Caspase-3明显降低,E2、AMH、Bcl-2明显升高(P <0. 05)。结论:针刺联合护卵汤可调节性激素和凋亡因子,促进卵泡生长,抑制卵泡闭锁,从而改善卵巢功能。
Objective:To observe the influence of combination of acupuncture and medicine on apoptotic factor and reproductive hormone of ovarian cells in rats with decreased ovarian reserve(DOR),and explore its mechanism of action.Methods:Seventytwo SD rats were randomly divided into the control group,the TCM group,the acupuncture group,the model group,the Western medicine group and the acupuncture group,the group of combination of acupuncture and medicine,with 12 rats in each group.Except for the control group,rats in the other five groups were given Tripterygium Glycosides Solution for gavage to establish DOR rat model which meets the experimental requirements.The ones in the group of combination of acupuncture and medicine,and the TCM group were given Ovum-protecting Decoction by gavage,while the ones in the acupuncture group and the model group were given the same amount of saline by gavage.The ones in the Western medicine group were given Climen Solution by gavage.At the same time,the group of combination of acupuncture and medicine,and the acupuncture group were given acupuncture.The above operation lasted for 16 days.Then the levels of serum estradiol(E2),follicle-stimulating hormone(FSH),luteinizing hormone(LH)and anti-mullerian hormone(AMH)were detected by El ISA.The expression of apoptotic factors Bax,Caspase-3 and Bcl-2 in ovarian tissue was detected by PCR.Results:Compared with the control group,the expression of FSH,LH,Bax and Caspase-3 in the model group increased significantly,while the expression of E2,AMH and Bcl-2 decreased significantly(P<0.05).Compared with the model group,FSH,Bax,Caspase-3 in the acupuncture group,the TCM group,the group of combination of acupuncture and medicine,and the Western medicine group decreased significantly,while their E2,AMH and Bcl-2 increased significantly(P<0.05).Conclusion:Acupuncture combined with Ovum-protecting Decoction can regulate sex hormones and apoptotic factors,promote follicular growth and inhibit follicular atresia,thereby improving ovarian function.
