摘要
探讨非抗凝动物血提取原卟啉二钠的方法。将凝固动物血中的蛋白质水解,分离出血红素,制备原卟啉甲酯、原卟啉等中间产物,最终精制获得原卟啉二钠;并根据原卟啉二钠的光吸收特性,利用分光光度法对其进行定量检测。原卟啉二钠为红褐色结晶,可溶于水,不溶于氯仿、乙醚及丙酮,易溶于酸性溶液。溶于酸性溶液后,在波长600nm、556nm、及408nm显示吸收峰,最大吸收波长为408nm。利用本方法可将非抗凝动物血精制成原卟啉二钠,且该方法是目前以非抗凝动物血为原料提取原卟啉二钠较为理想的方法。
The purpose of the authors is to find out an ideal method of extracting protoporphyrin disodium from inanticoagulant animal blood. Protoheme was separated from protein hydrolysate of coagulated animal blood and protoporphyrin and protoporphyrin methyl ester and other intermediate products were extracted, which were finally made into protoporphyrin disodium. According to its light absorption characteristics, protoporphyrin disodium was quantitatively detected. After determination, the extract of henna crystal was considered to be protoporphyrin disodium, which was soluble in water and insoluble in chloroform, Ether and acetone are easily soluble in acid solvent. When the extract in acid solvent was determined by spectrophotometer, absorptive peak occurred in 600nm, 556nm and 408nm wavelength, and the largest absorptive wavelength was 408nm. Protoporphyrin disodium can be extracted from inanticoagulant animal blood through the above method, which is an ideal method for protoporphyrin disodium extracted from inanticoagulant animal blood at present.
