dsRNA-RT-PCR技术在甜菜坏死黄脉病毒属不同分离物鉴定中的应用

摘要 从新疆甜菜主产区采集的 60份具丛根病症状的根、叶标样 ,经分离、纯化获得 7个代表性病毒分离物 .上述分离物分别人工接种昆诺阿藜 ,显症后 ,提取人工接种的昆诺阿藜患病组织的 ds RNA,RT-PCR检测表明 ,7个分离物均为甜菜坏死黄脉病毒 ( BNYVV) .ds RNA-RT-PCR技术具有灵敏度高、稳定性强及操作简便、安全等优点 ,可有效地完成对甜菜坏死黄脉病毒属 ( Benyvirus)不同分离物的鉴定 .其应用在国内尚属首次报道 . representative isolates of beet necrotic yellow vein virus(BNYVV)were obtained from 60 samples with the various symptoms by single spot isolation and purification from sugar beet main productious area of Xinjiang.Using dsRNA extracts of infected tissues as template for the reverse transcriptase polymerase chain reaction(RT-PCR),7 isolates were identified as BNYVV.Because of high sensibility,stability and easy and safe manufacture of this technique,it is used effectively in the identification of Benyvirus.Its application is first reported in China.
