摘要
目的:建立HPLC-ELSD法测定银杏内酯B脂微球中溶血磷脂酰胆碱的含量。方法:采用硅胶色谱柱(Platisil Silica,250mm×4.6mm,5μm),流动相为甲醇-冰醋酸(500∶10,三乙胺调节pH值至6.0),流速为1.0mL·min^(-1),柱温为40℃,检测器为蒸发光散射检测器。结果:溶血磷脂酰胆碱在38.4~480μg·mL^(-1)时,线性关系良好(r=0.9987),平均回收率(n=9)为102.13%,RSD为3.46%。结论:该方法准确,重现性好,可用于银杏内酯B脂微球中溶血磷脂酰胆碱的含量测定。
Objective:To establish an HPLC method for determination of lyso-phosphatidycholine in Ginkgolide B lipid microsphere. Methods:Chromatographic column was silicone column(Platisil Silica, 250 mm×4.6 mm,5μm),mobile phase was methanol-glacial acetic acid(500∶10, adjusting pH to 6.0 with triethylamine), the flow rate was1.0 mL·min-1, the column temperature was set at 40℃, and the detector was ELSD. Results:Lyso-phosphatidycholinehad a good linear relationin the rang of 38.4 ~480μg·mL-1(r=0.9987).The average recovery was 102.13%with the RSD of 3.46%(n=9). Conclusion:The method is accurate, reproducible and applicable for determination of lyso-phosphatidycholine in Ginkgolide B lipid microsphere.
