高效液相色谱法测定饲料中的盐酸氯丙嗪 被引量:2

摘要 盐酸氯丙嗪为抗精神分裂症药物,用于控制精神病患者躁动不安异常兴奋的状态。作用于动物时,能够使动物保持安静,使之处于睡眠或半睡眠状态。采用高效液相色谱法用岛津LC-10AT型高效液相色谱仪(紫外检测器),使用Eclipse SB-C18(5μm,4.6mm×150mm)色谱柱,流动相为乙腈∶0.02mol/L乙酸铵∶三乙胺(65∶35∶0.5)(用乙酸调pH=6.2),流速0.6 mL/min,柱温30℃,检测波长256nm。分析了饲料中盐酸氯丙嗪的含量,用甲醇提取饲料中的盐酸氯丙嗪,用该法测试杂质峰较少,且样品峰与杂质峰能较好的分离。通过精密度和加标回收实验检验方法的稳定性、可靠性都很良好。此法的检出限为0.02μg/mL,线性相关系数r2=0.9986,平均回收率为96.24%。 Chlorpromazine hydrochloride is an antipsychotic drug,which is used to control the restless or over-excited conditions of psychiatric patients.Being used on animals,it can keep the animals quiet and in a sleep or semi-sleep state.The high performance liquid chromatography(HPLC)method was used to determine the content of chlorpromazine hydrochloride in feed.Chlorpromazine hydrochloride in feed samples was extracted by methanol solution and analyzed using GS LC-10 AT HPLC(UV detector).The analytical column was Eclipse SB-C18(5μm,4.6 mm×150 mm).The mobile phase was acetonitrile∶0.02 mol/L ammonium acetate∶triethylamine(65∶35∶0.5)(pH was adjusted to 6.2by acetic acid).The flow rate of the mobile phase was 0.6 mL/min,and the column temperature was 30℃.The detection wavelength was 256 nm.By adopting this approach,less impurity peaks emerged,and a better separation between impurity peaks and analyte peaks appeared.The stability and reliability were proved to be good through precision and spike recovery experiments.The detection limit of this method was 0.02μg/mL.The method gave a good linear correlation coefficient(r2=0.9986),and the average analytical recovery was 96.24%.
