简便可见吸收光谱法测定药物中格列齐特的含量

摘要 在酸性三羟甲基氨基甲烷-盐酸介质中,格列齐特与溴甲酚绿反应生成具有1个正吸收峰和1个负吸收峰的离子缔合物,最大正吸收波长位于444 nm,最大负吸收波长位于614 nm,表观摩尔吸光系数(κ)分别为3.55×104 L·mol-1·cm-1(444 nm)和1.25×104 L·mol-1·cm-1(614 nm)。当以614 nm为参比波长,444 nm为测定波长,用双波长可见吸收光谱法测定时,表观摩尔吸光系数可达4.80×104 L·mol-1·cm-1。格列齐特在0~3.9 mg/L范围内服从朗伯-比尔定律。以灵敏度相对最高的双波长法为例测定市售格列齐特药物中格列齐特的质量浓度,加标回收率为98.18%~102.8%,相对标准偏差(RSD)为2.1%~2.5%(n=5)。 In the acidic trihydroxymethyl aminomethane-hydrochloric acid buffer media,gliclazide can react with bromocresol green to form a binary ion association complex with a positive(444 nm)and a negative(614 nm)absorption peak in the visible region.Their apparent molar absorptivities(κ)are 3.55×104 L·mol-1·cm-1(444 nm)and 1.25×104 L·mol-1·cm-1(614 nm),respectively.When 614 nm was selected as reference wavelength and 444 nm was selected as measured wavelength,the double wavelength visible absorption spectrometry was used to determine the concentration of gliclazide,and its apparent molar absorptivity(κ)can be achieved as 4.80×104 L·mol-1·cm-1.Gliclazide obeys Lombard Beer’s law in definite mass concentration range of 0~3.9 mg/L.The dual-wavelength method was applied to determine the content of glizide in commercially available glizide-containing drugs with a standard addition recovery and RSD(n=5)are in the range of 98.18%~102.8%and 2.1%~2.5%,respectively.
