芫花花蕾抗生育有效成分的研究——新的二萜原酸酯芫花酯丁的分离与结构

STUDIES ON THE ANTI-FERTILITY CONSTITUENT OF THE FLOWER YUAN-HUA ISOLATION AND STRUCTURE OF YUANHUATINE

芫花为瑞香科植物(Daphae genkwa Sieb.et Zucc.).前人曾从该植物的花、根部分得一系列抗生育有效成分——芫花酯甲(yuanhuacine)、芫花酯乙(yuanhuadine)、芫花酯丙(yuanhuafine)、12-苯甲酰氧基瑞香毒素(12-benzoxydaphnetoxin,genkwadaphnin,1)等.我们从芫花花蕾中也分到1.最近又分得一种新的二萜原酸酯化合物,命名为芫花酯丁(yuanhuatine,2).根据波谱分析并与1对比,确定其结构为1的1,2-双氢化合物.

A new diterpene orthoester,yuanhuatine,C_(34)H_(36)O_(10),m/z:604(M^+),[α]_D^(130)+112.1°(c0.390,GHCl_3),white amorphous powder,has been isolated in 0.0047% yield fromthe alcoholic extract of the flower of Yuan-Hua(Thymelaeaceae,Daphne genkwa Sieb.et Zucc.).The structure of yuanhuatine was shown to be 2 according to the spectralanalysis and in comparison with the known compound 1.The abortifacient minimumdose of 2 in monkey was shown to be 50μg/animal and LD_(50)=3.02±0.12mg/kg.
