肠道病毒71型3D蛋白潜在毒力位点的序列分析 被引量:2

Potential virulence-related mutations of 3D protein of enterovirus 71 based on sequence analysis

摘要 目的分析不同临床严重程度感染者肠道病毒71型的3D蛋白氨基酸序列,了解潜在毒力相关变异位点。方法从NCBI数据库下载不同临床严重程度C4亚型EV71感染者3D区核苷酸序列;对推导的氨基酸序列进行比对和分析,查找不同临床严重程度病毒感染者在3D蛋白存在差异有统计学意义的氨基酸位点。结果共下载序列113株,其中来自于重症及死亡患者序列共67株、轻症患者序列46株。分析显示病毒3D区的256位和370位氨基酸残基,在不同临床严重病例中的分布差异有统计学意义,其中部分重症患者在256位为非异亮氨酸,部分轻症患者在370位为非甘氨酸。结论 C4亚型EV71病毒的3D区256位和370位氨基酸的变异,可能与病毒毒力相关。 Objective To analyze 3D protein amino acid sequences of enterovirus 71(EV71)in patients with different clinical severity and explore potential virulence-related mutation.Methods The sequences of 3D in patients with different clinical severity were downloaded from NCBI database.The deduced amino acid sequences were aligned and statistical analyzed to find statistical significant substitutions.Results Totally 113 sequences were downloaded,in which 67 sequences from severe or fatal patients and 46 from mild patients.The residual distribution of amino acid 256 and amino acid 370 in 3D protein showed significant difference in patients with different clinical severity.Some severe cases were non-isoleucine at amino acid 256,and some mild cases were non-glycine at amino acid 370.Conclusion The variation of amino acid 256 and amino acid 370 of 3D protein may be related to virulence of subgenotype C4EV71.
