L-精氨酸对大鼠残余肾慢性肾脏纤维化进展的影响(英文)

The effect of L-arginine on the progression of chronic renal scarring in remnant kidney

摘要 目的 研究L 精氨酸 (L arg)对 5 / 6肾大部切除 (SNx)大鼠早期残余肾代偿性增生 (CRG)、小管间质细胞外基质成分 (ECM)堆集、长期生存率及肾脏纤维化的影响。方法 实验包括四组 (每组动物 5只 ) :(1)假手术组 ;(2 )假手术 +L arg组 ;(3)肾大部切除组 ;(4)肾大部切除 +L arg(L arg1%浓度经饮水中给予 )。大鼠分别于 30、12 0天处死。 30天实验组观察早期肾脏代偿性增生及小管间质免疫组化表达细胞外基质成分和α平滑肌肌动蛋白 (αSMA)。 12 0天实验组观察L arg对大鼠长期生存率及肾脏纤维化的影响。结果 L arg显著增加 (SNx)大鼠残余肾代偿性增生。SNx +L arg组小管间质增殖细胞核抗原 (PCNA)免疫组化表达较SNx组显著增加。而SNx +L arg组肾小球体积较SNx组增加了 16 %。L arg治疗显著增加肾大部切除大鼠小管间质胶原Ⅲ、Ⅳ、纤维连接蛋白的表达 ,而且此与小管间质α SMA表达呈显著正相关。在长期实验组 ,L arg治疗组生存率显著降低 (P <0 0 5 ) ,而肾脏纤维化程度明显加重 (P <0 0 5 )。结论 本研究提示早期给予肾大部切除大鼠L arg可能加重大鼠残余肾的肾脏纤维化程度。 OBJECTIVE: To investigate the effect of L-arginine (L-arg) on early compensatory renal growth (CRG), tubulointerstitial accumulation of extracellular matrix (ECM), long term survival rate and renal scarring in rats with 5/6 nephrectomy (SNx). METHODS: The experiment included four groups of rats (n = 5 each group): (1) Sham group, (2) SNx group, (3) SNx + L-arg group, and (4) Sham + L-arg group (L-arg 1% in drinking water). Parameters related with CRG and early tubulointerstitial expression of ECM and alpha smooth muscle actin (alphaSMA) were evaluated by immunohistochemistry at day 30.
