摘要
目的研究大鼠正畸复发过程中组织蛋白酶K抑制剂odanacatib对牙周组织改建的影响。方法 30只SPF级雄性大鼠,种植支抗60 g力近中移动右上颌第一磨牙,3周后拆除加力装置,建立复发模型。随机分为两组,每组15只。实验组右上颌第一磨牙牙周局部注射60μl、1.25μM odanacatib,每3天注射一次。对照组注射等量生理盐水。复发0天和复发2周时取大鼠上颌牙列模型,计算牙齿复发率。2周后处死大鼠,扫描Micro-CT,量化右上颌第一磨牙根分叉处骨矿物质密度(BMD)和骨体积分数(BVF)值,分析骨改建活性。TRAP染色分析两组破骨细胞数量的差异。免疫组化观察CatK及IGF-Ⅰ的表达变化。结果实验组复发率(42.45±0.11)%,小于对照组的(79.99±0.17)%,差异有统计学意义(P<0.05)。实验组的BMD和BVF大于对照组,差异有统计学意义(P<0.05)。两组破骨细胞数量无统计学差异(P>0.05)。实验组CatK的积分光密度(IOD)值小于对照组,差异有统计学意义(P<0.05)。实验组IGF-Ⅰ的表达高于对照组,差异有统计学意义(P<0.05)。结论局部注射odanacatib能有效地减少大鼠正畸后复发率,促进牙周组织改建,增加牙周组织的稳定性。
Objective To study the effect of cathepsin K inhibitor odanacatib on periodontal tissue remodeling during orthodontic relapse in rats.Methods Thirty male SPF rats were selected,and the right maxillary first molar were subjected to 60 g force and moved to mesially.After 3 weeks,the force device was removed and a relapse model was established.They were randomly divided into two groups.In the experimental group,60μl and 1.25μM odanacatib were injected into the periodontal of the right maxillary first molar,once every 3 days.The control group was injected with the same amount of saline.The rat maxillary dentition model was taken at 0 days and 2 weeks under relapse,and the relapse rate was calculated.After 2 weeks,the rats were sacrificed,and Micro-CT was scanned to quantify the BMD and BVF values of the right maxillary first molar root bifurcation,and the bone remodeling activity was analyzed.TRAP staining was used to analyze the difference in the number of osteoclasts in the two groups.The expression changes of CatK and IGF-I were observed by immunohistochemistry.Results The relapse rate of the experimental group was(42.45±0.11)%,which was less than that of the control group,(79.99±0.17)%.The difference was statistically significant.The BMD and BVF of the experimental group were larger than those the control group,and the difference was statistically significant.There was no significant difference in the number of osteoclasts between the two groups.The IOD value of the experimental group CatK was smaller than that of the control group,and the difference was statistically significant.The expression of IGF-I in the experimental group was higher than that in the control group,and the difference was statistically significant.Conclusion Local injection of odanacatib can effectively reduce the relapse rate after orthodontic treatment in rats,promote the reconstruction of periodontal tissues and increase the stability of periodontal tissues.
