摘要
目的研究成纤维细胞生长因子受体3对人恶性黑色素瘤A375细胞的调控作用。方法 2015年1月—2017年12月,收集郑州大学第一附属医院和山东大学省立医院恶性黑色素瘤组织及其癌旁组织样本46对,利用定量PCR、CCK-8等技术检测FGFR3对A375细胞增殖的和上皮间充质转化的调控。结果本研究中观察到过表达FGFR3能促进A375细胞的迁移和侵袭能力,而沉默FGFR3可以抑制其细胞增殖和侵袭能力。另外,FGFR3通过ERK、AKT和EGFR信号通路调控A375细胞的增殖和上皮间充质转化。结论成纤维细胞生长因子受体3是恶性黑色素瘤发生发展中的一个重要决定基因。
Objective To investigate the effects of FGFR3 on melanoma A375 cells.Methods To study the regulatory effect of fibroblast growth factor receptor 3,the expression of it was evaluated by immunohistochemical staining;malignant melanoma tissues and adjacent tissues were collected to detect the regulation of fibroblast growth factor R3 on the proliferation and epithelial mesenchymal transformation of A375 cells.Results We observed that FGFR3 was overexpressed in In this study,we observed that overexpression of this type of gene can promote the migration and invasiveness of cancer cells,while silencing this gene can inhibit cell proliferation and reduce its invasiveness.In addition,this gene can regulate the proliferation,migration and transformation of A375 cells through ERK signaling pathway.Conclusion Fibroblast growth factor receptor 3 is an important determinant gene in the development of malignant melanoma.
