摘要
以琼脂糖、纳米Fe_3O_4等为原料,采用反相悬浮再生法制得磁性琼脂糖微球,并偶联苄胺功能配基.采用X射线衍射、红外光谱、扫描电子显微镜等方法对微球进行表征.结果表明,微球球形度好,尺寸大小均一,平均粒径为60μm.包裹的Fe_3O_4含量约为10%,呈反式尖晶石结构.将微球用于分离牛血清蛋白(BSA),pH 5.0下对BSA的最大吸附容量为79mg/g,反复使用5次吸附容量降至原来的76%,表明微球吸附性能良好.
Using agarose,nano-Fe_3O_4 as the raw materials,magnetic agarose microspheres were prepared by the inverse suspension method and coupled with benzylamine ligand.The particles were characterized by using X-ray diffraction,infrared spectrum and scanning electron microscope.The results showed that the microspheres were spherical and possessed the average diameter of 60μm.The Fe_3O_4 packed in the microspheres was trans-spinel,with total content of about 10%.The microspheres were used to separate bovine serum albumin.Under pH 5.0,the maximum adsorption capacity was reached 79mg/g and the adsorption capacity decreased slightly to 76% after five cycled uses,indicating good adsorption performance.
