摘要
目的观察独活寄生汤对膝关节骨性关节炎模型大鼠NF-κB信号通路的作用,探讨其缓解膝关节骨性关节炎的可能机制。方法将大鼠随机分为空白组、模型组、纳曲酮组和独活寄生汤组,采用寒冷刺激法建立膝关节骨性关节炎模型。各组给予相应的药液灌胃。观察大鼠膝关节关节软骨病理形态变化;ELISA检测大鼠血清中IL-1、TNF-α的表达;Real time-PCR和Western bolt检测软骨中IκBα、NF-κBp65的基因和蛋白表达水平。结果纳曲酮组和独活寄生汤组大鼠的膝关节肿胀程度、活动度、饮食和精神都要优于模型组。纳曲酮组和独活寄生汤组大鼠膝关节软骨表面较粗糙,稍有不平整,软骨细胞排列较规则,炎性细胞浸润明显好转。与空白组比较,模型组大鼠血清中IL-1、TNF-α表达水平升高(P<0.01);与模型组比较,纳曲酮组和独活寄生汤组大鼠血清中IL-1、TNF-α表达下降(P<0.01,P<0.05)。与空白组比较,模型组大鼠软骨中IκBα、NF-κBp65的基因和蛋白表达水平升高(P<0.01);与模型组比较,纳曲酮组和独活寄生汤组大鼠软骨中IκBα、NF-κBp65的基因和蛋白表达水平降低(P<0.01,P<0.05)。结论独活寄生汤能缓解膝关节骨性关节炎模型大鼠的症状,其机制可能与其调节大鼠软骨中NF-κB信号通路的表达有关。
Objective To observe the effect of Duhuo Jisheng decoction on NF-κB signaling pathway in knee osteoarthritis(KOA)model rats,and to explore the possible mechanism of Duhuo Jisheng decoction in alleviating KOA.Methods Rats were randomly divided into blank group,model group,naltrexone group and Duhuo Jisheng decoction group.The model of KOA was established by cold stimulation.Each group was given corresponding liquid by gastric perfusion.The pathological changes of articular cartilage of knee joint in rats were observed;the expression of IL-1 and TNF-αin serum was detected by ELISA;the expression of genes and proteins of IxBa and NF-KBp65 in cartilage was detected by Real time-PCR and Western blot.Results The degree of knee joint swelling,range of motion,diet and spirit of rats in naltrexone group and Duhuo Jisheng decoction group were better than those in model group.In naltrexone group and Duhuo Jisheng decoction group,the cartilage surface of knee joint was rough,slightly out-of-flatness,the arrangement of chondrocyte was regular,and the infiltration of inflammatory cells was obviously improved.Compared with blank group,the serum levels of IL-1 and TNF-αin model group increased Objective To observe the effect of Duhuo Jisheng decoction on NF-B signaling pathway in knee osteoarthritis(KOA)model rats,and to explore the possible mechanism of Duhuo Jisheng decoction in alleviating KOA.Methods Rats were randomly divided into blank group,model group,naltrexone group and Duhuo Jisheng decoction group.The model of KOA was established by cold stimulation.Each group was given corresponding liquid by gastric perfusion.The pathological changes of articular cartilage of knee joint in rats were observed;the expression of IL-1 and TNF-in serum was detected by ELISA;the expression of genes and proteins of I B and NF-Bp65 in cartilage was detected by Real time-PCR and Western blot.Results The degree of knee joint swelling,range of motion,diet and spirit of rats in naltrexone group and Duhuo Jisheng decoction group were better than those in model group.In naltrexone group and Duhuo Jisheng decoction group,the cartilage surface of knee joint was rough,slightly out-of-flatness,the arrangement of chondrocyte was regular,and the infiltration of inflammatory cells was obviously improved.Compared with blank group,the serum levels of IL-1 and TNF-in model group increased(P<0.01),while the serum levels of IL-1 and TNF-in naltrexone group and Duhuo Jisheng decoction group decreased(P<0.01,P<0.05).Compared with blank group,the gene and protein expression of I B and NF-Bp65 in the cartilage of model group were increased(P<0.01).Compared with model group,the gene and protein expression levels of I B and NF-Bp65 in cartilage of the rats in naltrexone group and Duhuo Jisheng decoction group were decreased(P<0.01,P<0.05).Conclusion Duhuo Jisheng decoction can relieve the symptoms of KOA model rats,and its mechanism may be related to its regulation of the expression of NF-B signaling pathway in rat cartilage.
