苍术炒焦后氯仿部位增量成分初步研究 被引量:4

Preliminary Study on the Incremental Components of Chloroform in Atractylodes lancea L. after Stir-frying

摘要 目的观察苍术炒焦后氯仿部位的增量成分,并初步建立其含量测定方法。方法对苍术炒焦前后高效液相色谱法(HPLC)色谱图进行比较,确定苍术炒焦后氯仿部位增量明显的色谱峰;进一步分离纯化、鉴定增量化合物结构;采用高效液相色谱法测定确定的增量成分含量,色谱条件为Agilent C18(5 m,4.6mm×250mm)色谱柱,流动相为乙腈-水(5∶95),检测波长284nm,柱温25℃,流速1.0mL/min,进样量为10L。结果苍术炒焦后5-羟甲基糠醛等7个成分含量显著增加,主要增量成分之一确定为5-羟甲基糠醛(5-HMF);测定道地产区6个不同批次的苍术及焦苍术片中5-HMF的平均含量分别为0.0057%,0.105%,其结果表明苍术炒焦后5-HMF的含量是生苍术的15倍以上。结论苍术炒焦后增量成分中5-HMF增加极显著,该结果为阐释苍术炒焦后药效变化提供了物质基础支撑。 Objective To investigate the incremental components of chloroform in Atractylodes lancea L.after stirfrying,and initially establish its content determination method.Methods The chromatographic peaks of chloroform were determined by comparing the chromatograms of Atractylodes lancea L.before and after stir-frying.The structures of the compounds were further separated,purified and identified.The content of the incremental components was determined by HPLC with Agilent C18(5 micron,4.6 mm×250 mm)column flowing.The phase was acetonitrile-water(5:95),the detection wavelength was 284 nm,the column temperature was 25℃,the flow rate was 1.0 mL/min,and the injection volume was 10 mL.Results The content of 5-hydroxymethyl furfural(5-HMF)and other seven components increased significantly after stir-frying.One of the main incremental components was 5-HMF.The average content of 5-HMF in six different batches in real production area of Atractylodes lancea L.and Jiaozhang tablets was 0.0057%and 0.105%respectively.The results showed that the content of5-HMF after stir-frying was more than 15 times of raw Atractylodes lancea L..Conclusion The increase of 5-HMF in the incremental components of Atractylodes lancea L.after stir-frying is very significant,which provides a material basis for explaining the changes of pharmacodynamics of Atractylodes lancea L.after stir-frying.
