摘要
比较了不同基本培养基,不同激素及其不同浓度组合对番茄愈伤组织的增殖、不定芽分化及其生根的效果.结果表明:MS+NAA0.02~0.10mg·L-1+6-BA3.0mg·L-1+蔗糖30g·L-1(pH=5.8)均可诱导愈伤组织的增殖,但以MS+6-BA3.0mg·L-1+NAA0.05mg·L-1+蔗糖30g·L-1最佳;MS+IAA0.1mg·L-1+6-BA2.0mg·L-1+蔗糖30g·L-1培养基对愈伤组织分化不定芽的效果最佳;MS+IAA0.1mg·L-1+蔗糖20g·L-1培养基对生根较好.
The effects of different media with various hormone concentrations on tomato callus inducing, shoot differentiation, and rooting were compared. It showed that callus could be induced in MS media with 3.0 mg·L-1 of 6-BA plus 0.02-0.1 mg·L-1 of NAA and 30 g·L-1 of sugar(pH5.8). The best medium was MS with 3.0 mg·L-1 of 6-BA plus 0.05 mg·L-1 of NAA and 30 g·L-1 of sugar, in which callus grew fast and many embryo calli were produced. 2 basic media appending different concentrations of 6BA and IAA, IBA or NAA were used for shoot differentiation from calli. It demonstrated that MS media with 2.0 mg·L-1 of 6-BA plus 0.1 mg·L-1 of IAA and 30 g·L-1 of sugar were the best to bud differentiation, in which shoots were strong as well as less variation. The best rooting medium was MS with 0.1 mg·L-1 of IAA and 20 g·L-1 of sugar.
