槲皮素共振光散射法测定微量蛋白质

摘要 研究了槲皮素与蛋白质在pH=4.20条件下作用的共振光散射特征,并以此建立了测定微量蛋白质的新方法。考察了影响因素,确定了最佳反应条件。对牛血清白蛋白,测定的线性范围为0—5.0mg/L。检测限57.5μg/L。方法用于人血清试样中总蛋白的测定,结果与经典的考马斯亮兰法一致。 Resonance light scattering(RLS) characteristics of interaction between quercetin with proteins in acidic medium of pH4.2 was studied. The effects of interference substances and optimization of procedure were investigated and a new method for the determination of the proteins serum albumin (BSA) is 0 - 5 .0mg/L and limit of detection is 57 . 5μg/L.The method was applied to the determination of total proteins in human serum sample and the results were compared with the traditional proteins assay (CBBG - 250 method) .
