摘要
目的 建立快速检测风疹病毒特异性IgM抗体的改良ELISA法。 方法 利用聚乙二醇 (PEG )能加速抗原抗体反应 ,促使其沉淀的特点 ,在常规间接ELISA法的一抗和二抗中加入 4%的PEG ,以缩短反应时间。结果 用该法测定 39份风疹阳性血清和 183份阴性血清的风疹病毒IgM抗体 ,阳性血清检出率为 97.4% ,与常规间接ELISA检测结果 (92 .3% )一致 ;阴性血清的检出率为 95 .1% ,高于间接ELISA(87.5 % ) ,经配对资料的 χ2 检验 ,差别有统计学意义。结论 在风疹IgM间接ELISA法稀释液中加入PEG ,可以缩短试验时间 。
Objective To establish an improved ELISA for the rapid detection of rubella IgM antibody.Method 4% PEG,which can accelerate the antigen antibody reaction and actuate their deposition,was added in the dilution of the primary antibody and the HRP labeled antibody of normal indirect ELISA,to shorten the reactive time.Results Using the improved ELISA to detect the rubella IgM antibody in 39 rubella serum specimens and 183 normal serum specimens,the positive rate was 97.4%,which was coincident with the result of indirect ELISA(92.3%);and the negative rate was 95.1%,which was significantly higher than that of indirect ELISA(87.5%).Conclusions Adding PEG in the dilution of rubella IgM in indirect ELISA can shorten the reactive time,and promote the accuracy of the detection.
