植物激素在条斑紫菜原生质体固体培养中的作用 被引量:11

SOME EFFECTS OF PHYTOHORMONE IN THE SOLID CULTURE OF PORPHYRA YEZOENSIS PROTOPLASTS

摘要 利用酶解技术制备高活力的条斑紫菜原生质体;通过优化原生质体的制备工艺和培养条件,建立了条斑紫菜原生质体的无菌培养体系。同时首次证明了高等植物组织培养中常用的两种激素NAA和6一BA对紫菜细胞生长具有明显地促进作用,并应用这两种激素建立了条斑紫菜原生质体的固体培养方法。为开展紫菜遗传转化中阳性克隆的定向筛选,以及探讨紫菜的基础生理生化反应机制奠定了良好基础。 Vegetative protoplasts of porphyra yezoensis are prepared with the sea snail enzyme, and axenic culture of protoplasts is accomplished following optimizing the preparing process and culture conditions. For the first time, the two phytohormones, NAA (α—Naphthaleneacetic acid ) and 6-BA (6-Benzyladenine) have been shown to have remarkable stimulative growth effects on protoplasts of porphyra yezoensis (Fig6,Tablel), which nave been applied effectively, in higher plant tissue culture, and we established a solid culture method forit (Figl). the new results lay a good foundation for directional screening of positive clones in P.yezoensisgenetic engineering,meanwhile it provides valuable resesrch tools to elucidate physiological responses or biochemical control mechanisms in P. yezoensis.
