摘要
目的 探讨多西紫杉醇在细胞和器官水平抗血管生成的作用和机制。方法 采用四唑盐比色实验(MTT)、内皮细胞迁移和小管形成实验、大鼠动脉环培养等方法检测药物对人脐静脉内皮细胞 (HUVEC)的增殖、迁移、小管形成和大鼠动脉环血管生成的作用。以上实验均分为空白对照组和多西紫杉醇的溶剂对照、0 .0 5、0 .1、0 .2、0 .5、1.0ng/mL 7个组。 结果 ①各组MTT的平均吸光度值分别为 0 .3 0 7± 0 .0 7、0 .3 0 5±0 .0 8、0 .3 0 1± 0 .0 5、0 .2 98± 0 .0 6、0 .2 5 1± 0 .0 4、0 .2 2 4± 0 .0 5、0 .2 0 5± 0 .0 2 ;②每高倍镜下发生迁移的细胞数量各自为 44 .3 6± 4.8、42 .2 8± 5 .2、3 7.75± 4.6、2 8.65± 3 .6、15 .5 9± 2 .1、10 .82± 1.3、7.43± 0 .6;③平均小管形成长度依次为 192 .3 6± 11.5 4、188.89± 7.5 9、175 .5 3± 11.5 4、166.78± 12 .3 5、15 1.62± 16.3 8、118.65± 11.5 8、78.93± 3 .67μm ;④大鼠动脉环第 13天新生血管数目依次为 10 1.0 0± 10 .8、97.88± 7.8、73 .0 0± 6.6、60 .88±5 .8、5 2 .2 5± 5 .3、3 3 .13± 2 .8、18.13± 1.3。以上 4项实验的空白和溶剂对照、MTT实验的前 4组之间的两两比较均不存在显著差异 (P >0 .0 5 ) ,其余各组间两两比较?
Purpose. To study the antiangiogenesis effect of docetaxel on cell and organ levels. Methods. MTT experiment, endothelial cell migration and capillary formation experiment and rat aortic ring culture methods were adopted to evaluate the effect of docetaxel on Human umbilial vein endothelial cell (HUVEC) function of proliferation, migration, capillary formation and neovascularization of rat aortic ring. All experiments were classify into blank, docetaxel solvent control, 0.05, 0.1, 0.2, 0.5, 1.0ng/mL groups. Results. 1 The mean MTT A value were 0.307 ± 0.07, 0.305 ± 0.08, 0.301 ± 0.05, 0.298 ± 0.06, 0.251 ± 0.04, 0.224 ± 0.05, 0.205 ± 0.02 respectively; 2 the migrated number cell of above groups were 44.36 ± 4.8, 42.28 ± 5.2, 37.75 ± 4.6, 28.65 ± 3.6, 15.59 ± 2.1, 10.82 ± 1.3, 7.43± 0.6 respectively; 3 the mean capillary length of them were 192.36 ± 11.54, 188.89 ± 7.59, 175.53 ± 11.54, 166.78 ± 12.35, 151.62 ± 16.38, 118.65 ± 11.58, 78.93 ± 3.67 μm respectively; 4 number of new capillaries sprouted of each rat aortic ring group on 13th day were 101.00 ± 10.8, 97.88 ± 7.8, 73.00 ± 6.6, 60.88 ± 5.8, 52. 25 ± 5.3, 33.13 ± 2.8, 18.13 ± 1.3 respectively. There were no significant differences between the blank and solvent groups above, as well as blank, docetaxel solvent control, 0.05, 0.1 ng/mL group of MTT results (P > 0.05), there were significant differences between other groups (P < 0.01). Conclusions. With or without cytotoxic role, docetaxel has inhibitory effect on proliferation, migration, capillary formation function of HUVEC and neovascularization of rat aortic ring, it is a promising antiangiogenic.
