致痫大鼠海马神经元氨基酸类物质含量变化的研究 被引量:16

摘要 目的 研究癫痫发病中海马神经元氨基酸类物质的合成、分泌情况 ,探索癫痫发病的可能机制。方法 用马桑内酯致痫培养大鼠海马神经元细胞 ,用高效液相色谱法测定培养神经元细胞内外谷氨酸 (Glu)、天门冬氨酸 (Asp)、γ 氨基丁酸 (GABA)、甘氨酸 (Gly)的含量。结果 致痫海马神经元细胞Glu合成相对增加 ,分泌显著性增加 (P <0 0 1) ;Asp合成没有明显增加 (P >0 0 5 ) ,或相对减少 ,分泌显著性增加 (P <0 0 1) ;GABA分泌显著性减少 (P <0 0 1) ,合成相对地减少 ;Gly合成分泌显著性减少 (P <0 0 1)。结论 癫痫发病中 ,痫性神经元细胞合成和 Objective To study the synthesizing and secretion of hippocampus neuron amino acid on epilepsy, and explore the pathogenesis of epilepsy attack.Methods We treated culturing hippocampus neuron cell in rats by coriaria lactone(CL), and measured the contents of glutamate(Glu), asparte(Asp), gama aminobutyric(GABA) and glycocine(Gly) of the internal and external culturing hippocampus neuron cell by high performance liquid chromatography(HPLC).Results The Glu synthesizing of epilepsy hippocampus neuron cell increased relatively, the Glu secretion increased significantly( P <0 01). The Asp synthesizing didn't increased significantly( P >0 05), or decreased relatively, the secretion increased significantly( P <0 01). The GABA secretion dereased significantly( P <0 01), the GABA synthesizing decreased relatively. The Gly synthesizing and secretion dereased significantly( P <0 01).Conclusion The abnormal of the Glu, Asp, GABA, Gly synthesizing and secretion may be the base of neurotransmitter balance disorder in epilepsy.
