双层活性皮肤替代物的实验研究

摘要 目的探索活性皮肤替代物体外培养的途径。方法常规分离皮肤角质形成细胞和成纤维细胞,用I型 胶原作为真皮基质,采用气-液界面方式进行培养,用HE染色观察培养物的结构。结果培养的皮肤替代物具有近似 皮肤的形态结构,表皮部分有角质层形成。结论本方法可用于活性皮肤替代物的制备。 Objective To establish a method of constructing a bilayered living skin equivalent in vitro. Methods Using serially passaged keratinocytes and dermal fibroblasts with a matrix of type I collagen,living skin equivalent (LSE) has been generated through air-liquid interface culture. Sections of LSE were stained with hematoxylin and eosin and examined under microscopy. Results Morphology of LSE was similar to that of skin. A protective cornified layer was developed. Conclusion The method employed here can be used to generate living skin equivalent.
