胃癌细胞系Lewis抗原表达和体外诱发免疫应答反应

Expression of Lewis blood group antigen and in vitro study of immune response induced in human gastric cancer cell lines

目的 :探讨由于幽门螺旋杆菌 (Helicobacterpylori,H .pylori)和人胃黏膜上皮表达相同的Lewis抗原 ,在宿主抗H .pylori的免疫应答中 ,发生自身免疫反应的可能性。 方法 :采用流式细胞术和乳酸脱氢酶 (lactatedehy drogenase,LDH)释放试验 ,在检测 3种胃癌细胞系Lewis血型抗原表达状况的基础上 ,进行体外抗原抗体结合诱发抗体依赖性细胞介导的细胞毒性效应实验。结果 :KATOⅢ、80 3和 82 3细胞系均有Lewis抗原的表达 ,但各自的表型和量不同。在体外诱发抗体依赖性细胞介导的细胞毒性效应中 ,除 80 3细胞系anti H .pylori和anti Leb 组外 ,其余 3种细胞系各种抗体组均有不同水平的LDH的释放。结论 :初步提示Lewis抗原抗体结合诱发自身免疫反应存在的可能性 。

Objective: To explore the possibility of autoimmune reaction induced by the mimicry of Lewis antigen between H.pylori and host infected. Methods: Flow cytometry and LDH release assay were used to screen the expression of Lewis antigen in 3 human gastric cancer cell lines and study antibody dependent cell mediated cytotoxicity (ADCC) induced in vitro , respectively. Results: Lewis antigens were expressed in KATOⅢ, 803 and 823 cell lines differently and LDH was released in 3 gastric cell lines at different levels in ADCC induced in vitro . Conclusion: Our results suggested that the mimicry of Lewis antigen might induce autoimmune reaction in host immune response against H.pylori infection, which needs further study and verification.
