摘要
目的观察CD_3AK细胞系的超微结构及其与杀伤功能相关的特征。方法用透射电镜观察有代表性的细胞的超薄切片。结果胞膜较完整,有少量突起,外形不规则,周围未见支原体污染。胞质中有膜泡分泌颗粒;大量游离核糖体,少量脂滴和线粒体;未见溶酶体。有的细胞胞质和细胞器较少。核形状不规则,常有凹陷,核染色质聚集成大块状。结论所建CD_3AK细胞系透射电镜下的超微结构具正常T淋巴细胞的特征,胞质中有与杀伤靶细胞相关的颗粒结构。
Objetive To explore ultrastructure of the established CD3AK cell line and its characteristics related to killing activity. Methods Ultrathin sections of representative cells were observed with transmission electron microscope.Results CD3AK cells were of irregular outline and more intact cytoplasm membrane with procrusions, without infection of Mycoplasmas around the cell.In cytoplasm, there were some membrane bubble secreting granules, lots office ribosomes, a few of mitochondria and lipid drops, and no lysosomes to be seen.Some cell had less cytoplasm and fewer organdies. Cell nucleus, which had depression, was irregular in appearance, and had aggregating chromatin.Conclusions Under the transmission electron microscope, the ultrastructure of the established CD3AK cell line is characterized by Tlymplocytes.there are some granules ultrastructure related to killing target cells in cytoplasm.
