摘要
目的 研究错配修复基因hMSH3和hMSH6在散发性结直肠癌发生中的作用。方法 应用放射性同位素为基础的PCR技术 ,检测了 4 8例散发性结直肠癌和 2 0例癌旁组织中hMSH3和hMSH6基因的突变。结果 hMSH3和hMSH6在散发性结直肠癌中的突变率分别为 5 / 4 8(10 .4 % )和12 / 4 8(2 5 % ) ,在癌旁组织中未见突变。
Objective To study the effect of hMSH3 and hMSH6 in the development of sporadic colorectal cancer. Methods We used radioactive polymerase chain reaction (PCR) to study the mutation of mismatch repair gene hMSH3 and hMSH6 in 48 surgical specimens of sporadic colorectal cancer and 20 specimens of adjacent tissue of tumor. Results The mutation of hMSH3 and hMSH6 occurred in 10.4%, 25% of the sporadic colorectal cancer specimens respectively, didn't occure in adjacent tissue of tumor specimens. Conclusions This results indicate that mutations of hMSH3 and hMSH6 play an important role in the development of sporadic colorectal cancer.
