摘要
来氟米特 (leflunomide ,LFM )是一种化学合成的小分子新型免疫抑制剂。本文研究了LFM对实验性自身溶血性贫血的防治作用和与环孢素 (CSA )的协同作用。结果表明 ,2 2 5mg/kg小鼠的LFM可抑制小鼠产生抗自身红细胞抗体滴度上升 ,能有效预防自身溶血性贫血的发生 ,使自身抗体的滴度维持在 1∶16左右。小剂量的LFM与亚治疗剂量的CSA合用具有良好的协同作用。 4 5mg/kgLFM与 10mg/kgCSA合用使自身抗体滴度不超过 1∶48左右 ,其CI值为 0 75 ;9mg/kgLFM与 10mg/kgCSA合用使自身抗体滴度维持在 1∶4左右 ,其CI值为 0 2 3。
Leflunomide (LFM) is an isoxazole derivative which has shown some new immunomodulating properties. The present study was carried out to investigate the effects of domestic LFM on the experimental mouse autoimmune hemolytic anemia and its synergic effect with ciclosporin (CSA). LFM at a dose of 22 5mg/kg (p.o) could inhibit the production of anti erythrocyte autoantibodies and prevent autoimmune hemolytic anemia. When two low doses of LFM 4 5mg/kg and 9mg/kg were respectively combined with a subtherapeutic dose of CSA 10mg/kg, the autoantibody titres of the two combined groups were about 1∶48 and 1∶4. The combination index were 0 75 and 0 23 respectively. These autoantibody titres are much lower than those of the single drug used alone.
