摘要
目的研究阻断转化生长因子-β(TGF-β)受体的信号转导对正常皮肤成纤维细胞增殖和I型胶原合成的调控。方法组织块法体外培养皮肤成纤维细胞,加入缩短型TGF-βⅡ型受体的重组腺病毒(实验组,50 pfu/cell)或β-半乳糖苷酶重组腺病毒(对照组,50 pfu/cell),培养后行细胞计数和Western Blot检测细胞增殖率和I型胶原合成状况。结果 缩短型TGF-βⅡ型受体在皮肤成纤维细胞的过度表达能有效抑制细胞增殖达34%~50%,并显著减少I型胶原的合成。结论过度表达缩短型TGF-βⅡ型受体可以通过阻断TGF-β的信号转导来抑制成纤维细胞的细胞增殖和I型胶原合成。
Objective To study the effect of blocking TGF-β signaling on dermal fibroblasts proliferation and type I collagen production. Methods In vitro cultured dermal fibroblasts were treated with recombinant adenovirus containing a truncated type II TGF-β receptor gene (50 pfu/cell) in experimental group or β-galactosidase viruses (50 pfu/cell)in control group. Their effects on cell proliferation and type I collagen production were analyzed by cell counting and Western blot respectively. Results Over-expression of truncated TFG-β receptor II in dermal fibroblasts inhibited cell proliferation ( 34 %-51 % ) and type I collagen production. Conclusion Over-expression of truncated TGF-β receptor II gene in dermal fibroblast can decrease cell proliferation and type I collagen production via blocking TGF-(3 receptor signaling.
