摘要
目的综述端粒酶的组成、结构及其活性调节机制。方法回顾近几年国内外端粒酶研究的最新进展。结果端粒酶是一多亚基复合物,具有逆转录酶活性。它能延长端粒,维持端粒的正常功能,在生殖细胞、干细胞、绝大多数肿瘤细胞、永生化细胞株等细胞中活化,所以端粒酶与肿瘤、细胞衰老有密切关系。结论端粒酶活化和活性调节机制为肿瘤的诊断、治疗和预后提供了新的思路,它已成为肿瘤研究的新靶点。
Objective:To explore the composition,struction and activity regulative mechanism of telomerase.Methods:To review the up to date progresses of telomerase in recent years throughout the world.Results:Telomerase is a reverse transcriptase comprised of many subunits.It can elongate telomere and maintain the normal functions of telomere,and be activated and examined in reproductive cells,stem cells,as well as the majority of tumor cells and immortalizational cells line.So telomerase is strong related to cancer and cellular senescence.Conclusions:Activation and activity regulative mechanisms of telomerase in tumor cells have provided a new apporach for tumor's diagnosis,theraph and prognosis.Telomerase is regarded as new target of tumor research.
