摘要
目的 :探讨培养软骨用作生长板移植替代物的可行性。方法 :自 1月龄兔分离软骨细胞 ,离心管培养 2周形成软骨。切取 6周龄兔胫骨生长板 ,与培养软骨进行组织学、胰岛素样生长因子Ⅰ型受体α链 (IGF ⅠRa)免疫组织化学、3H 胸腺嘧啶脱氧核苷 ( 3H TdR)掺入率检测和电镜观察。结果 :培养软骨具有一定的骺软骨组织学结构 ,缺乏典型的生长板软骨细胞柱状排列。培养软骨和生长板IGF IRa免疫组织化学染色均为阳性。培养软骨3H TdR掺入率显著高于生长板 (P <0 .0 1)。培养软骨中细胞外基质结构疏松 ,5 %软骨细胞呈现凋亡。结论 :培养软骨具有一些与生长板相似的性质 ,可能成为生长板的移植替代物。
Objective:To compare cultured cartilage with growth plate and investigate possibility of using cultured cartilage as a substitute for growth plate.Methods:Chondrocytes were isolated from articular cartilage surface of rabbit aged 1 month and formed cartilage after cultivation for 2 weeks.Tibial growth plate was obtained from rabbit aged 6 weeks.Both growth plate and cultured cartilage were histologically observed.The expression of insulin-like growth factor Ⅰ receptor α(IGF-ⅠRa) were immunohistologically detected and the proliferation of chondrocytes was detected by 3H-TdR.The comparison of cultured cartilage and growth plate was also performed by transmission electronic microscope.Results:Cultured cartilage histologically formed some characteristics of epiphyseal cartilage,but it lacked typical arrangement like column of chondrocytes in growth plate.Immunohistological staining of IGF-ⅠRa was positive in cultured cartilage and growth plate.The incorporation rate of 3H-TdR in cultured cartilage was markedly higher than that in growth plate(p<0.01).The structure of extracellular matrix of cultured cartilage was scattered and 5% of chondrocytes showed apoptosis.Conclusion:Cultured cartilage possess some characteristics like growth plate,and may be used as a graft substitute for growth plate.
