摘要
目的 探讨NMDA诱导神经元胆囊收缩素mRNA表达及其机制。方法 利用体外培养的胚胎大鼠大脑皮层神经元和NorthernBlot技术 ,观察NMDA诱导神经元胆囊收缩素mRNA的表达以及c -fos、c - jun反义寡聚核苷酸对该作用的影响。 结果 NMDA刺激神经元 2h后 ,胆囊收缩素mRNA的表达量与对照相比升高 317% (P <0 0 1) ,而预先给予 5 0uMol~ 10 0uMol的c -fos、c - jun反义寡聚核苷酸后该作用被明显抑制 (抑制率分别为 71 5 %、2 16 5 % ,P <0 0 1)。结论 NMDA可通过c -fos、c - jun的介导作用诱导神经元合成胆囊收缩素 。
Objective To investigate the mechanism on expression of CCK mRNA NMDA-induced in cultured neurons of rats Methods The cerebral cortex neurons primary cultured of rat's embryo were utilized Expression of CCK mRNA NMDA induced in cultured neurons and effects of antisense oligonucleotides of c-fos and c-jun on it were detected with northern blot Results The expressions of CCK mRNA in cultured neurons significantly enhanced when cultures were exposed to NMDA for 2 hours and that were inhibited (71 5% and 216 5%, P<0 01) by antisens oligonucletides of c-fos and c-jun(50uMol~100uMol) Conclusion These results indicated that effect of NMDA inducing expression of endogenous CCK mediated by c-fos and c-jun many be associated with neuroprotection of NMDA
