摘要
目的 :研究丁酸钠逆转人卵巢癌 3AO细胞恶性表型的机制。方法 :采用MTT法观察药物对卵巢癌细胞的抑制作用 ,扫描电镜观察药物作用后细胞超微结构变化 ,免疫组化染色观察药物作用后转移抑制基因KAI1表达的变化。结果 :药物对卵巢癌 3AO细胞的抑制率呈剂量依赖性 ,药物作用后细胞微绒毛减少、变短、消失 ,KAI1基因免疫组化染色较对照组明显增强。结论 :丁酸钠可成功逆转人卵巢癌细胞的恶性表型 。
Objective:To study the mechanism of sodium butyrate (NaB) on human ovarian cancer cell line 3AO.Method:The inhibiting effect of sodium butyrate on cell line was studied by MTT colormetric assay,the morphologic changes by electron microscope,and variation of KAI1 gene expression by immohistory chemistry.Results:The inhibiting rate of cell was dependented on amount of sodium butyrate (NaB).The surface villi were lessen,shorten and disappeared.Expression of KAI1 gene was significently stronger than control.Conclution:Sodium butyrate can reverse the malignant characteristics of ovarian cancer cell line.The KAI1 gene may be important in tumor metastasis.
