摘要
目的 :探讨正脂丸对体外培养的人血管内皮细胞产生一氧化氮 (NO)、内皮素 (ET)的影响 ,以阐明正脂丸保护内皮和抗动脉粥样硬化的作用机制。方法 :用血清药理学方法制备不同浓度的正脂丸含药血清 ,并以该血清作用于体外培养的人血管内皮细胞 ,分别用硝酸还原酶法和放射免疫法检测内皮细胞培养液中NO、ET的含量。结果 :正脂丸可以促进体外培养的内皮细胞生成NO ,且具有剂量依赖关系 ,各组间两两比较 ,差异有显著性 (P <0 .0 1) ;同时可以抑制ET的生成 ,与对照组比较 ,差异有显著性 (P <0 .0 1)。结论 :正脂丸可以调节内皮细胞产生NO、ET ,这可能是其保护内皮和抗动脉粥样硬化的作用机制之一。
Objective:To investigate the effects of Zhengzhi Pill(ZZP) containing serum on the productions of nitric oxide(NO) and endothelin(ET) by endothelial cells.The endothelial protective effect and anti atherosclerosis effect of ZZP was explored to elucidate the pharmacological mechanism of ZZP.Method:Various density Zhengzhi Pill(ZZP) containing sera were prepared.Endothelial cells were cultured with those sera and the levels of NO and ET in supernatants were detected respectively.Result:ZZP increased NO production and simultaneously inhibited ET expression(P<0.01).Conclusion:ZZP has anti atherosclerosis endothelial protective effects.The mechanism maybe relate to its regulating the synthesis of NO and ET in endothelial cells.
