摘要
目的 研究伤寒杆菌耐药质粒 pRST98与细菌外膜泡 (OMV)形成的关系。方法 将 ( 1)携带 pRST98的野生型耐药伤寒杆菌———ST1及ST2 ;( 2 )不含 pRST98的野生型敏感伤寒杆菌———ST3及ST4 ;( 3 )不含质粒的大肠杆菌———E .coliK12 W14 85;( 4)用十二烷基硫酸钠消除 pRST98的突变体菌株———ST1(R-)及ST2 (R-) ;和 ( 5 )经接合转移将pRST98导入敏感型伤寒杆菌及大肠杆菌的接合子———pRST98/ST3、pRST98/ST4 及pRST98/E .coliK12 W14 85用于实验 ,用透射电镜观测细菌在有无 pRST98时 ,OMV形成的差异。结果 携带 pRST98的耐药伤寒杆菌消除 pRST98前后 ,不含 pRST98的敏感伤寒杆菌和大肠杆菌在导入该质粒前后 ,细菌形成OMV的数量和形态均无明显变化。
Objective To explore the relationship between drug resistant Salmonella.typhi R plasmid (pR ST98) and outer membrane vesicles (OMV) of bacteria.Methods Following strains were used in the experiment:(1) plasmid-containing wide-type S.typhi strain ST 1 and ST 2,(2) plasmid-free sensitive wide-type S.typhi strains ST 3,ST 4,(3) plasmid free sensitive E.coli K 12W 1485,(4) plasmid-cured strain ST 1(R -),ST 2(R -),(5) plasmid-introduced sensitive strains pR ST98/ST 3,pR ST98/ST 4,pR ST98/E.coliK 12W 1485.Their ability of OMV formation was compared by transmission electron microscope.Results The outer membrane vesicles of plasmid-free bacteria and those of bacteria harbouring pR ST98 were not found to be significantly different.Conclusion In vitro pR ST98 can not affect the ability of bacteria to form outer membrane vesicles.
