以从黑龙江省腹泻犊牛粪便中分离的牛轮状病毒(bovine rotavirus)为抗原,免疫6~8周龄BALB/_c小鼠,取血清抗体阳性小鼠的脾细胞与SP_(2/0)骨髓瘤细胞融合。用ELISA间接法检测抗体,通过有限稀释法,克隆出一株分泌抗牛轮状病毒单克隆抗体的杂交瘤细胞株。经鉴定,此杂交瘤细胞株分泌的单克隆抗体为IgM,轻链为K链。通过连续传代证明,此杂交瘤细胞株具有稳定分泌单克隆抗体的性质。杂交瘤细胞诱生BALB/_c小鼠腹水的单克隆抗体滴度可达1:10000以上。
BALB/_C mice were immunized with bovine rotavirus (BRV), which was isolated from the feta of calves with diarrhea in Heilongjiang Province. The spleen cells taken from the mice with serum positive to BRV were fused with SP_(2/0)cells. After the antibodies to BRV were detected with the indirect ELISA, one strain of hybridoma cells secreting anti-BRV McAb was clonized by the limited dilution. This McAb was IgM with K light chain. The hybridoma cell strain could secrete the anti-BRV McAb steadily. The titre of the antibodies induced by the hybridoma cells in the abdominal water was more than 1: 10000.
