摘要
目的 :探讨脊髓损伤对局部骨组织中BMP、IL -6、TNF -α表达变化的影响并讨论其在骨代谢变化中的作用。方法 :通过横断大鼠胸髓建立脊髓损伤的动物模型 ,对脊髓损伤后 1、6周时股骨近段松质骨中BMP ,IL -6,TNF -α进行免疫组织化学染色 ,并结合计算机图像分析系统对表达变化进行评定。结果 :脊髓损伤后 1周时TNF α阳性表达强度显著高于对照组 (P <0 .0 5 ) ,6周时与对照组相比差异无显著性。BMP表达在 1周时与对照组差异无显著性意义 ,6周时表达增强显著 (P <0 .0 5 ) ,IL -6表达在伤后 1周与对照组比较差异无显著性 ,伤后 6周高于对照组 ,差异有显著性意义 (P <0 .0 5 )。结论 :TNF -α早期增加 ,BMP和IL -6在晚期增加。BMP、IL -6、TNF -α是影响脊髓损伤后骨代谢变化的重要因素。
Objective:To explore the changes of BMP,IL 6 and TNF α protein expression in trabecular bone of rats with spinal cord injury(SCI)and its significance in bone matabolism.Methods:Experimental model of SCI was eatablished by resecting spinal cord at the tenth thoracic vertebrae in SD rats.BMP,IL 6 and TNF α proteins were examined by using immunochemical stain at 1,6 weeks postoperatively.The intensity of BMP,IL 6 and TNF α were determined with the use of imaging analysis techniques.Results:The expression of TNF a in SCI group rose at week 1 significantly( P <0.05),and TNF α expression in SCI group returned normal at week 6 compared with that in control group.Immunochemistry stain of BMP and IL 6 were more intensive in SCI groups at week 6 than that in control groups( P <0.05,0.05,respectivelly),however,no difference was found between two groups at week 1.Conclusion:The expression of TNF α increasing in the early stage and BMP,IL 6 rising lately indicate that cytokines play a critical and different role in bone metabolism after SCI.
