摘要
目的 :探讨非生物标本的电镜样品制备及超薄切片技术。方法 :选择无色透明纤维、黑色纤维及无色透明薄膜等材料 ,进行脱水、环氧树脂包埋、超薄切片。结果 :切出满意切片 ,并在电镜下拍摄出高质量的照片。结论 :根据非生物标本的特性成功地进行了电镜样品制备及超薄切片 。
Objective: To investigate the technology in preparation and ultramicrotomy of non-biological specimens for electron microscopy.Methods:Dehydration,epon embedding and ultra-thin sectioning were carried out with black fibers, colorless hyaloid fibers and membranes.Results:Good quality ultra-thin sections and fine electron micrographs were produced. Conclusion:The preparation of non-biological specimens and their ultramicrotomy were successfully carried out according to their characteristics,which made it possible to study the ultrastructure of non-biological specimens.
