摘要
目的 :观察树突状细胞 (DC)捕获抗原后的移行、抗原提呈方式及其在抗原特异性皮炎中的作用。方法 :利用异硫氰酸荧光素 (FITC)的可显示性 ,将其作为抗原皮内免疫CBA小鼠 ,测定局部淋巴结的淋巴细胞、DC总数并作淋巴细胞增殖试验 ,用FACS分析DC携带抗原。结果 :皮肤经FITC刺激后 ,局部淋巴结的树突状细胞及淋巴细胞数明显高于对照组 (P <0 .0 5 ,P <0 .0 1) ;淋巴细胞增殖试验亦有明显差异 (P <0 .0 5 ) ;FACS分析的结果表明 ,局部淋巴结内的树突状细胞百分率及荧光强度均明显提高 ;荧光显微镜观察呈现不均一的膜荧光 ,提示该细胞抗原提呈方式为膜结合型。经病理切片观察 ,再次注射抗原的局部皮肤明显有单核、淋巴细胞浸润为主的皮肤迟发型超敏反应的病理变化。结论 :FITC所引起的抗原特异性皮炎是一种细胞介导的迟发型超敏反应 ,DC在致敏过程中起了重要的作用。
Objective:To observe the function of Dendrtic cells(DC) on uptake antigen,migration antigen presentation and the pathlogical role in antigen-specific cutaneous inflammation.Methods:Flurescein isothiocyanate (FITC) was injected into mice's intraderme.The numbers of DC and lymphocytes were assayed.The state of DC-born specific antigen was observed.Results:The results showed that DC migrated to the regional lymphnodus after uptake the antigen.There appeared an obvious increasing in the numbers of Dendrtic cells and lymphocyte cells (P<0.05 P<0.01)isolated from the local lymphnodus.The results of lymphocyte proliferation test in both specific antigen (FITC) showed significant difference and conA were higher than those in the control group (P<0.05).The numbers and fluorescent density increased by FACS analysis of DC.The fluorescein could be found on the surface of DCs with fluorescence microscopy.Conclusion:We conclude that the antigen-specific cutaneous inflammation is the cell-mediated hypersensitivity reaction,in which DC plays an important role in the sensitivity process.
