摘要
目的探索骨髓基质细胞源神经干细胞对脑损伤的治疗作用。方法将犬自体骨髓基质细胞在体外扩增并经BrdU标记及预分化处理后,通过脑室或循环途径将其植入脑干损伤模型组及对照组动物脑内或体内,并通过组织学方法观察其在损伤脑区及其它脑区的分布情况。结果骨髓基质细胞源神经干细胞自体移植治疗组(C、D组)BrdU阳性细胞在脑损伤区的分布明显多于(P<0.05)对侧非伤区、对照组(A组及B组)同部位、及其它非损伤脑区。结论骨髓基质细胞源神经干细胞对损伤脑区有亲合性;骨髓基质细胞源神经干细胞可通过向损伤脑区的迁移而发挥治疗作用。
Objective To explore the action of bone marrow stromal cells (BMSCs) in treatment of brain injury following NSCs transplantation. Methods After BMSCs were proliferated, marked with BrdU and pre-differentiated in vitro, the BrdU-marked BMSCs-derived-NSCs were transplanted into animals brains or bodies in the experimental and control groups through brain ventricle or vein systems, and then their distributions were observed in varied brain areas histologically. Results The amount of BrdU positive cells within damaged areas in the auto-transplantation groups (group C and group D) were significantly larger than that in the symmetrical brain area and the other brain areas in the same group as well as in the same brain area in control groups (group A and group B) (P<0.05). Conclusion The BMSCs-derived-NSCs have the affinity to injured brain areas. They exert the therapeutic action on brain stem injuries by way of immigrating to the injured brain areas.
