青心酮与孕妇血清蛋白结合率的实验研究

Serum Protein -binding Rate of Qingx ingtong in Pregnant Women

目的比较青心酮在正常和早、晚孕妇血清中的蛋白结合率及相关参数的差异。方法应用平衡透析法了解青心酮在早、晚孕妇血清中的蛋白结合率,并用HPLC色谱法进行定量检测,对结合率及相关参数进行比较。结果青心酮与正常妇女血清的蛋白结合率为(73.69±10.96)%,在早孕妇女血清蛋白结合率为(73.17±9.31)%,在晚孕妇女血清的蛋白结合率为(60.93±7.06)%。青心酮与1%人血清白蛋白的结合率为(42.96±17.57)%。青心酮在平衡时与血清蛋白的结合常数(Kp)为1.4766×10-3mol/L,结合位点(n)为0.17,在正常妇女血清的白蛋白结合药物的表观最大能力(Bp)为3.69μmol/g,早孕妇女为5.34μmol/g,晚孕妇女为2.14μmol/g。在正常妇女血清的药物-复合物的解离常数(Kdp)为27.36μmol/L,早孕妇女为27.87μmol/L,晚孕妇女血清为12.36μmol/L。结论青心酮主要是与血清中白蛋白结合。早、晚孕妇血清中的蛋白结合率及结合参数的差异与血清中的蛋白含量相关。

Objective To compare the serum protein -bindin g rate of Qingxintong in non -pregnan t women,early Preg-nant women and late Pregnant women.Methods The protein -binding rate of Qingxin tong was estimated by protein equili-bration dialyzator and was determin ed by HPLC.Results The protein binding rate of Qingxint ong was(73.69±10.96)%,(73.17±9.31)%and(60.93±7.06)%in non -pregnant women,early Pregn ant women and late Pregnant women respectively.The binding rate of Qingxintong with 1%albumin was(42.96±17.57)%.The association constant in equilibrium was 1.477×10 -3 mol /L,and the binding site was 0.17.The maximum binding power of drug -albumin complex in non -pregnant women,early Pregnant women and late Pregnant wo men was 3.69μmol /g,5.34μmol /g and2.14μmol /g respectively,and the dissociation constant of the complex was 27.36μmol /g,27.87μmol /g and 12.37μmol /g respectively.Conclusion Qingxintong binds with serum albumi n chiefly.The significant differen ce of protein binding rate and parameters in early and late Pregnant women is correlate d to the serum protein level.
