重组绵羊白细胞介素2基因的分泌性表达及表达产物的提取

Expression of Ovine Interleukin-2 Gene and Purification of Expressed Products

将构建的绵羊白细胞介素2基因表达菌株(PMPOIL2)接种在营养肉汤培养基上,培养20h,分别收集菌体及上清液。菌体超声裂解;上清液中的蛋白质分别用MgCl2、(NH4)2SO4、NaCl沉积。用抗Pilin-IL2血清检测菌体裂解物及上清沉淀物,结果上清沉淀物中有凝集抗原,而菌体中没有,并且(NH4)2SO4沉积的效果最好。

The strain(PMPOIL2) expressing ovine Interleukin-2 was inoculated into broth culture medium and cultured 20h ,the bacterium and the supernatant were collected.The bacterium was split by supersonic,and protein in the supernatant was precipitated with MgCl2,(NH4)2SO4 and NaCl.The specific reaction of the recombinant ovine Interleukin-2 and antiserum of Pilin-IL2 was demonstrated by cross electrophoresis in the precipitate of supernatant and was not in the bacterium,the effect was the best with (NH4)2SO4.
