摘要
应用特异于 HMG- box区域的兼并引物 ,以基因组 DNA为模板扩增了东北虎的 SOX基因 .在扩增产物中发现一条 2 2 0 bp的扩增带 .经过克隆、序列测定和同源性检索 ,发现一个基因片段与人、小鼠、鸡和爪蟾 Sox3的同源性分别为 95 %、95 %、92 %和 88% ;与人和小鼠 SRY基因的同源性分别为 75 %和 73% .因此该片段应当为东北虎的 Sox3基因片段 ,被命名为 Pt Sox3.另一片段与人、小鼠、爪蟾和大鼠 Sox4的同源性分别为 98%、97%、95 %和 94% ;与人和小鼠 SRY基因的同源性分别为 5 6 %和 5 4%被命名为 Pt
The SOX genes of Panthera tigris altaica Temminck were detected by a pair of degenerate primers. The degenerate primers were special to the SOX genes family within the HMG.box. With the genomic DNA as template, one specific band was observed with the length of 220bp. One fragment of DNA was founded with the identities to HSOX3, Msox3, Csox3 and XlSox3 95%,95%,92% and 88% respectively. The similarities of this fragment to HSRY and Msry were 75% and 73% respectively. Therefore, it was part of SOX3 in Panthera tigris altaica Temminck, and was named as PtSox3. The identities of another fragment to SOX4 of human, mouse, Xenopus laevis and rat were 98%,97%,95% and 94% respectively. The similarities of this fragment to HSRY and Msry were 56% and 54% respectively. So, it was named PtSox4.
