摘要 为更好地保存样本,提取高质量的基因组DNA,采取先将试验材料用75%乙醇固定,然后再提取基因组DNA的方法,经过检测后用于RAPD等基因工程分析。与从新鲜材料和液氮冷冻材料提取DNA方法的比较表明,用乙醇固定材料提取DNA是一种确实可行的方法,并克服了通常用新鲜材料或液氮冷冻材料所存在的不足(DNA容易降解)和远途异地取材的困难。 In order to search for a suitable method for tissue preservation and DNA extraction,the muscle of abalone was fixed with 75% ethanol firstly.The genomic DNA extracted successfully from the fixed tissue was tested by electrophoresis and Uvvisible spectrophotometer and was used in RAPD analysis.Compared with the DNA isolated from fresh tissue and frozen one with liquid nitrogen,the DNA extracted from fixed tissue is of high quality and high molecularweight.There is almost the same result in RAPD analysis with the DNA frozen samples.The research result suggested 75% ethanol was fit for tissue preservation and the tissue fixed with 75% ethanol was quite good for DNA extraction.In addition,the method could get rid of the difficulty in sample collection from some distant fields and could lower the test cost.
