摘要
目的 :探索白细胞精子症的白细胞的产生 ,是否与IL 2 ,IL 8和NO有关 .方法 :白细胞精子症患者 4 0例 (平均年龄 2 6± 4岁 ) ,精液中IL 2 ,IL 8水平测定采用双抗体夹心ELISA法 .白细胞密度采用邻甲苯胺 过氧化物酶染色法 .用镀铜镉还原荧光法检测NO代谢产物硝酸盐 (NO-3 ) .结果 :白细胞精子症不育组精液IL 2 ,IL 8和NO水平分别为 (2 0 .5± 5 .4 ) ,(6 .3± 0 .8) μg·L-1和 (10 8.6± 2 .0 ) μmol·L-1;明显高于非白细胞不育组 (11.8± 6 .7) ,(2 .4± 0 .4 ) μg·L-1和(75 .8± 2 .3) μmol·L-1及生育组 (11.4± 6 .8) ,(1.8± 0 .4 )μg·L-1和 (40 .8± 1.5 ) μmol·L-1,(P <0 .0 0 1) .非白细胞不育与生育组相比IL 2 ,IL 8水平无显著差异 (P >0 .0 5 ) ,NO有差异 (P <0 .0 1) .结论 :白细胞精子症不育精液IL 2 ,IL 8水平升高使男性生殖管壁中淋巴细胞活化 ,白细胞聚集于精液 .IL诱导NO合成对精子和生殖组织具有毒性和损害作用致男性不育 .
AIM: To investigate the distribution of interleukin 2 (IL 2), interleukin 8 (IL 8) and nitric oxide (NO) in the white blood cells (WBC) produced by leukocytospermia patients . METHODS: The levels of IL 2 and IL 8 in 40 patients were detected using ELISA method. The O toluidine/Goodpaetures staining was employed to check the concentration of WBCs, and the product of metabolism, nitrate (NO - 3) was determined with copper coated cadmium reductive fluorescence assay. RESULTS: The levels of IL 2, IL 8 and NO were (20.5±5.4), (6.3±0.8) μg·L -1 and (108.6±2.0) μmol·L -1 in leukocytospermia patients respectively and remarkably higher than those in non leukocytic infecundity patients (11.8±6.7) , (2.4±0.4) μg·L -1 and (75.8±2.3) μmol·L -1 ( P <0.001) and normal fertility group (11.4±6.8) , (1.8±0.4) μg·L -1 , (40.8±1.5) μmol·L -1 . There was no significant difference in the levels of IL 2 and IL 8 between non leukocytic infecundity patients and the normal group ( P >0.05) but the difference in the NO level between these two groups was obvious ( P <0.001). CONCLUSION: The levels of IL 2 and IL 8 that increase in leukocytic infecundity patients, activate the lymphocytes in the wall of spermatic tract, and make result in leukocytes accumulation in sperm. The synthesis of NO induced by IL is toxic and can damage the sperms and germ cells , leading to male infecundity.
