摘要
目的 :研究标准曲线法测定血清超氧化物歧化酶。方法 :应用催化褪色反应原理于邻苯三酚自氧化法测定超氧化物歧化酶 ,在pH8.3 0的KH2 PO4~Na2 B4O7缓冲溶液中邻苯三酚 (反应浓度 3mmol·L-1)自氧化反应 15min ,在波长 3 3 7nm处 ,对照管和样品管吸光度比值的对数与超氧化物歧化酶质量浓度成正比。结果 :超氧化物歧化酶质量浓度为 2 .2 5~2 7.0 0mg·L-1;标准曲线方程为 :lgA0 /AS=-0 .0 0 73 + 0 .0 12 3C ,r=0 .9995 ;血清加标样回收率均值为 10 0 % (s=3 .0 % ,n =2 4) ;日内和日间变异 (RSD)分别为 4.5 % (n =10 )和 5 .6%(n =4)。结论 :催化褪色反应原理可应用于血清超氧化物歧化酶的测定。
Objective To study the standard curve method for determination of superoxide dismutase (SOD) in serum.Method The principle of catalytic fading reaction was applied to the pyrogallol autoxidation for determination of SOD in serum.The pyrogallol of the reaction concerntration 3?mmol·L -1 was used for autoxidation 15?min in the pH8.30 KH 2PO 4~Na 2B 4O 7 buffer solution.The logarithm of the ratio( A 0/ A S),the absorbance A 0 of the reference tube to the absorbance A s of the sample tube at 337?nm was proportional to the concerntrations of SOD.Result The standard curve equation with the SOD concerntrations range from 2.25 to 27.0?mg·L -1 (the activity:from 3.25 to 39.00?U·ml -1 )was obtained as follows: lg A 0/ A S= -0.007?3+0.012?3 C , r =0.999?5.The mean recovery of 100%( s =3.0%, n =24)was found by the presented method.The relative standard deviations within the day and between days were 4.5% and 5.6% respectively.Conclusion The principle of catalytic fading reaction is successfully applied to the pyrogallol method for determination of SOD in serum.
