摘要
目的 :构建hIL 15 TNFα 融合毒素的表达质粒 ,为该融合毒素的功能研究及临床应用前景的分析评价奠定基础。方法 :将hIL 15成熟肽基因与人工改造的TNFα 基因按正确的阅读框架融合 ,定向克隆于表达载体pET16b。结果 :经酶切分析及DNA序列分析 ,所构建的质粒均插入hIL 15 TNFα 融合基因。结论 :利用pET16b表达载体可成功构建融合毒素hIL 15 TNFα
Objective To characterize the biological function and clinical perspective of hIL?15?TNF α fusion toxin,a plasmid expressing this fusion molecule was constructed.Method Artifically modified TNF α cDNA was fused precisly to the reading frame of hIL?15 and further cloned into the E.coli expression vector pET16b.Results The recombinant plasmid containing hIL?15?TNF α fusion gene was obtained as demonstrated by restriction analysis and DNA sequencing.Conclusion The recombinant plasmid expressing hIL?15? TNF α fusion toxin is successfully constructed.
