金雀异黄素对胃癌细胞iNOS表达影响与抑癌作用研究 被引量:2

摘要 目的 研究金雀异黄素 (Genistein ,Gen)对人胃腺癌细胞SGC 790 1增殖影响和对诱导型一氧化氮合酶 (inducednitricoxidesynthase ,iNOS)基因表达的影响。 方法 应用分裂指数法观察Gen抑制癌细胞生长 ,WesternBlot及RT PCR方法检测iNOS的表达情况。结果 Gen显著抑制了SGC 790 1细胞的增殖 ,并可显著诱导iNOSmRNA转录、增加iNOS蛋白表达 ,呈剂量效应和时间效应关系。结论 Gen抑制胃癌细胞增殖的机制之一可能与其上调iNOS基因表达有关。 Objective To study the effects of genistein on the proliferation and the iNOS expression of human gastric carcinoma SGC 7901cells.Methods Cell mitosis test was used to investigate the effect of genistein on cell proliferation. Western blot and RT PCR were used to observe the effects of Gen on iNOS gene expression.Results Genistein inhibited the proliferation of tumor cells significantly and increased iNOS mRNA transcription and protein expression in the dose dependent and time dependent manners.Conclusion Proliferatory inhibition of human gastric carcinoma cells after treated with genistein may be due to the up regulation expression of iNOS.
