摘要 目的:探讨利用裸鼠建立感染丙型肝炎病毒(HCV)体内动物模型的可能性。方法:将体外感染了HCV的人胎肝细胞(HFH)移植到裸鼠脾脏。移植术后3个月,取脾作病理学检查,以评估感染了HCV的HFH是否还存活。并且通过透射电镜检查,以评估移植入裸鼠脾脏的HFH胞浆中是否仍有HCV。同时,取裸鼠的外周血液作液相RT-PCR检查,以明确外周血液中HCV RNA是否为阳性。结果:移植术后3个月,移植入裸鼠脾脏内的HFH依然存活,经检查移植入裸鼠脾脏中的HFH胞浆内仍然存在HCV样颗粒。然而,体内有HCV感染的裸鼠外周血液仅70%HCV RNA为阳性。结论:裸鼠可作为“活试管”,支持体外感染了HCV的HFH在其脾脏内较长期生存。但仅使用外周血液HCV RNA检查这一个指标来判断裸鼠体内是否有HCV感染是不够的。 Objective: To explore possibility establishing the animal model with HCV infection in vivo by means of using nude mice. Methods: Human fetal hepatocytes (HFH) infected with HCV in vitro were transplanted into the spleens of nude mice. 3 months after transplanting, the spleens of the nude mice were taken out to carry out pathological examination to evaluate whether HFH infected with HCV still survived, and transmission electron microscopeexamination to evaluate whether there were still HCV in the cytoplasm of HFH transplanted into the spleens. Meanwhile, the peripheral blood of the nude mice was carried out liquid RT-PCR examination to make clear whether HCV RNA in the blood was positive. Results: 3 months after transplanting, HFH transplanted into the spleens of nude mice still survived, and HCV-like particles were still present in the cytoplasm of HFH in the spleens of nude mice. Nevertheless, the peripheral blood of the nude mice with HCV infection in vi-vo was only 70% positive for HCV-RNA. Conclusion: Nude mice may be used as 'live test tubes' to support HFH infected with HCV in vitro to survive for longer periods of time in their spleens. But it is insufficient that only using the index of HCV RNA examination in peripheral blood to judge whether nude mice have HCV infection in vivo.
