摘要
目的 :确定建立腹主动脉瘤动物模型时弹力蛋白酶最短灌注时间和腹主动脉最佳游离长度。方法 :80例大鼠随机均分为 1组生理盐水对照组和 7组弹力蛋白酶实验组。灌注时间分别为 10、2 0、30、6 0和 12 0min ,游离长度分别为 0 .5和 1.0cm。术后 14d观察各组大鼠腹主动脉直径扩张率及动脉壁的组织学变化。结果 :≥ 30min组腹主动脉直径平均扩张率均 >10 0 % ,与 10、2 0min及对照组有显著差异 (P <0 .0 5 ) ;而 30min组与 6 0、12 0min组无明显差异 (P >0 .0 5 )。≥ 30min各组的大鼠腹主动脉中弹力蛋白均严重受损 ,且与游离腹主动脉长度无关。结论 :在弹力蛋白酶诱导的大鼠腹主动脉瘤模型中 ,弹力蛋白酶灌注时间可由 12 0min降至 30min ,游离腹主动脉长度可从 1.0cm缩至 0 .5cm ,不影响腹主动脉瘤的形成。
Objective:This study was performend to determine whether it would be possible to shorten elastase infusion time and infused aortic length in the elastase induced rat aneurysm model.Methods:The abdominal aortas of 80 male Wistar rats were dissected and infused for 10,20,30,60 or 120min with a solution containing 50U of elastase or for 30min with saline solution(control)at the arotic length of 0.5 or 1.0cm.On day 14,the surviving rats underwent laparotomy.The diameter of the aorta was measured,and the aortic tissue was excised for histological examination.Results:The diameter of the ≥30 min elastase infused aortas was significantly larger than the diameter of the 10 min,20 min elastase infused and saline infused aortas(P<0.05).There were no significant differences between the 30 min and the 60 min and 120 min elastase infused aortas(P>0.05).Microscopically,there was total or subtotal loss of elastic tissue of ≥30 min elastase infused aortas.It has nothing to do with the infused aortic length.Conclusion: It is possibleto shorten the elastase infusion time from 120min to 30min and infused aortic length from 1.0cm to 0.5cm.
