氧化苦参碱抑制SMMC—7721细胞增殖的研究 被引量:23

摘要 目的 研究不同浓度氧化苦参碱对人肝癌细胞SMMC-7721增殖的抑制作用。方法SMMC-7721细胞经不同浓度氧化苦参碱处理后,用MTT法检测细胞生长和增殖,用流式细胞仪技术检测细胞DNA分布。结果氧化苦参碱浓度>2.5mg/ml,细胞毒作用较大;浓度在0.1~2.5mg/ml时,对肿瘤细胞的生长有抑制作用,且呈时间剂量依赖性;浓度<0.1mg/ml,对肿瘤细胞生长几乎无抑制作用。氧化苦参碱作用后可增加G0/G1期细胞所占的百分比,阻止细胞进入S期。2.5mg/ml氧化苦参碱作用于SMMC-7721细胞3d,可观察到凋亡峰。结论 一定浓度的氧化苦参碱有抑制SMMC-7721细胞增殖的作用。 Objective To study the proliferation inhibition of different concentrations of oxymatrine on human hepatoma cell SMMC-7721. Methods The SMMC-7721 cells were treated by oxymatrine in different concentrations and variable durations. Cell growth and proliferation were observed using tetrazolium dye (MTT) assay. Flow cytometry was used to assay cell-DNA distribution. Results When the drug concentration was higher than 2. 5mg/ml, the major effect was cell necrosis. Oxymatrine in the concentrations from 0. 1mg/ml to 2. 5mg/ml could inhibit proliferation of SMMC -7721 cells as revealed by MTT assay and the cell - survival rate curve. Increase in the percentage of G0/G1 phase cell was observed by flow cytometry, which suggested that the inhibition effect of oxymatrine on SMMC - 7721 cells is to block them at G0/G1 phase. In addition, the drug concentration of 2. 5mg/ml at 3 successive days on SMMC -7721 cells could be observed to induce apoptosis. Conclusion Oxymatrine could inhibit the proliferation of SMMC - 7721 cells.
