摘要
应用高效液相色谱仪、蛋白顺序仪和傅里叶变换离子回旋共振质谱仪等仪器 ,纯化并测试了公驼诱导排卵因子(OIF)的部分活性序列和降解过程。结果表明 ,公驼OIF是单链多肽 ,分为活性序列和相对稳定固定序列两部分 ,N 端的活性位序仅为 6个残基 :赖氨酸 缬氨酸 丙氨酸 苯丙氨酸 丝氨酸 苏氨酸。该因子的氧化降解过程在冰冻条件下是由氨基酸侧链基团的氧化和肽链的断裂共同引发的。
Partial polypeptide sequence and degradation process of the ovulation inducing factor (OIF) extracted and purified from seminal plasma in Camelus bactrianus were analyzed by the high performance liquid chromatography (HPLC),protein sequencer ABI 476A and the Fourier transform ion cyclotron resonance (FTICR) APEXⅡ.The results showed that OIF in camel seminal plasma is a polypeptide with single chain which consist of the bioactive sequence and the relatively stable squence, that the bioactive sequence in N terminus of OIF is K V A F S T, and that degradation process of oxidation in OIF under freezing condition occurr coincidently at side chain groupsoxidation withpeptide bonds breaks in amino acid.
