肾舒胶囊对人系膜细胞增殖及分泌肿瘤坏死因子-α和纤连蛋白的影响

EFFECT OF SHENSHU CAPSULE ON CELL PROLIFERATION AND SECRETION OF TUMOR NECROSIS FACTOR-α AND FIBRONECTIN IN CULTURED HUMAN MESANGIAL CELL

目的 :研究肾舒胶囊对人系膜细胞 (HMC)增殖及分泌肿瘤坏死因子 -α(TNF -α)和纤连蛋白 (FN)的影响。方法 :采用血清药理学方法 ,将肾舒胶囊含药血清加入培养的HMC中 ,MTT法测定HMC增殖情况 ,ELISA法测定HMC分泌TNF -α及FN情况。结果 :肾舒胶囊含药血清可明显抑制HMC增殖及其分泌TNF -α和FN ,且其抑制作用呈一定的浓度依赖性和时间依赖性。结论 :肾舒胶囊可能通过抑制MC增殖及其分泌TNF -α等炎症介质和FN等细胞外基质 (ECM)

Objective: To investigate the effect of ShenShu capsule on cell proliferation and secretion of tumor necrosis factor-α (TNF-α) and fibronectin (FN) in cultured human mesangial cell (HMC). Methods: Cultured HMC was treated with sera containing ShenShu capsule ingredients in different concentration, then MTT incorporation was used to assess the proliferation of HMC and ELISA assay was used to measure the secretion of TNF-α and FN. Results: Sera containing shenShu capsule ingredients could inhibit cell proliferation and TNF-α and FN secretion of HMC in a concentration-dependent and time-dependent manner. Conclusion: ShenShu capsule can inhibit cell proliferation and secretion of TNF-α and FN of HMC, which may be one of its mechanisms in the treatment of chronic renal failure (CRF).
